Phylogenomic analysis of the Porphyromonas gingivalis - Porphyromonas gulae duo: approaches to the origin of periodontitis

Porphyromonas gingivalis is an oral human pathogen associated with the onset and progression of periodontitis, a chronic immune-inflammatory disease characterized by destruction teeth-supporting tissue. P. belongs to genus Porphyromonas, which being composed Gram-negative, asaccharolytic, non-spore-forming, non-motile, obligatory anaerobic species, inhabiting niches such as cavity, urogenital tract, gastrointestinal tract infected wound from different mammals including humans. Among genus, stands out for its specificity in colonizing cavity keystone role periodontitis pathogenesis. To understand evolutionary process behind context Pophyoromonas this study, we performed comparative genomics study publicly available genomes, focused on four main objectives: (A) confirm phylogenetic position phylogenomic analysis; (B) definition comparison pangenomes relative gulae ; (C) evaluation gene family gain/loss events during divergence (D) pressure (represented calculation Tajima-D values dN/dS ratios) comparing families . Our analysis found 84 high-quality assemblies representing 14 strains (from total 233 genomes). Relevant virulence factors, FimA, Mfa1, hemagglutinins, conserved gulae, gingivalis, suggesting origin those factors occurred previous - divergence. These results suggest unexpected relationship between duo showing more clues about periodontitis.
